Does it make any difference if I create my translation engine in SQL Server 2000 or SQL Server 2005. Are there any advantages with SQL Server 2005 compared to SQL Server 2000?

No. Sisulizer uses very basic features of DB server. Both SQL Server are pretty identical in the basic features. We have tested and used SQL Server 2005 much more than 2000 Jaakko

Is it possible to move an existing TE from one server to another by doing backup and restore?
